2022 Workshops:
1. Basic introduction to X-ray computed tomography & 3D imaging
Presenter: Muofhe Tshibalanganda
Pre-recorded, will be available for questions after the video.
The workshop will cover the basics of X-ray Micro-CT and Nano-CT.
Course content will include:
· Specific examples
· How the method works
· What kind of information can be found
· Advantages of X-ray 3D imaging
Systems will be demonstrated and prepared data will be used for demonstrating 3D data analysis. This will cover basic 3D visualization, image segmentation, defect analysis, part to part comparison and many more. The course focuses on new users and is ideal for researchers and students new to the field of non-destructive "X-ray microscopy".

2. Water Isotope Analysis by Laser Spectroscopy

"The purpose of this course is to provide comprehensive hands-on training in the installation, operation and maintenance of laser spectrometry for the analysis of stable isotopes in water samples and the use of the LIMS for Lasers software.
The course comprises lectures covering theoretical concepts on the measurement of environmental stable isotopes using laser spectrometry, followed by demonstrations and practical exercises including the setup of the laser analyser, sample preparation, as well as a brief overview of isotope data postprocessing.

Hyperspectral imaging and chemometrics

The first video will provide an overview of the theory and principles of this technique to enable a better understanding of how NIR spectroscopy and hyperspectral imaging works. It will describe typical applications which make use of this instrument and technique. The second video will focus on an introduction to multivariate data analysis which can be used to explore trends in data and to develop qualitative and quantitative models.

10. An introduction to Serial Block-Face (SBF) Scanning Electron Microscopy (SEM)

Serial Block-Face (SBF) Scanning Electron Microscopy (SEM) is a powerful imaging modality that allows for 3D acquisition SEM samples at high resolution. This is of great value for biological research, as cells and tissues can be studied in their native 3D morphology which allows for a deeper understanding of cellular phenotypes associated with certain pathologies. When combined with other fluorescent microscopy techniques, SBF-SEM provides immense value for the localization and distribution of proteins, bacteria or viruses within tissues which can provide novel insights into certain disease states. This workshop will serve as an introduction to SBF-SEM and explain the workflow involved in sample preparation and image acquisition. Due to the large amount of data generated by this approach, an overview of data handling and image processing will also be given to introduce researchers to basic principles of machine learning in the era of 'Big Data'.

11. Introduction to biological sample preparation for SEM

Biological sample preparation is essential in scanning electron microscopy (SEM). Flawed sample preparations can undermine the quality of results and lead to false conclusions. Thus, the aim of this talk is to equip researchers and students with the essential knowledge required to prepare samples for SEM investigations in life sciences. To obtain acceptable SEM images with good ultrastructural preservation requires careful application of the SEM sample preparation method.

13. Introducing to biological sample preparation for STEM

We talk about the advantages of STEM with the availability of various types of STEM techniques, different detectors and a variety of adjustments in order to suit specific samples and analysis requirements. 19. Correlative Light and Electron Microscopy
Presenters: Lize Engelbrecht and Jurgen Kriel
The workshop on correlative light and electron microscopy will show the participant how two microscopy modalities, confocal/super-resolution microscopy and scanning electron microscopy, can be used on the same sample to exploit more information from the sample than would have been possible with any of the two modalities separately. The benefits of this technique and the different workflows to achieve correlation will be discussed, but also the inherent challenges of the technique. We will also introduce an open source software package we use for ultimate correlation.

20. Super-resolution

In this workshop, the participants will be introduced to the latest developments in the field of super-resolution microscopy techniques. The ability of microscopes to resolve detail in fluorescence images at sub-resolution limits is becoming more and more accessible to biological researchers. In the workshop we will explain in more detail also the specific aspects of SR SIM and SMLM, two of the super-resolution techniques we can perform here in Stellenbosch.
